Advancement in the field of molecular biology and technical engineering has improved the sequencing methods and results for NGS-based monitoring and diagnostic test market. NGS platforms enable various methods for the analysis of genome, transcriptome or epigenome of an organisms. Variation in the sequencing methods differ by obtaining the DNA or RNA samples and data analysis options which are used to analyze the results.
The main concept behind NGS technology is similar to Sangers sequencing. In the latter, DNA polymerases catalyzes the incorporation of fluorescently labeled deoxyribonucleotide triphosphates (dNTPs) into a DNA template at the time of sequential cycles of DNA synthesis. During each cycle, the nucleotides are identified by fluorophore excitation at the point of incorporation. The major difference is that rather than sequencing a single DNA fragment, NGS sequences millions of fragments parallelly.
The genomic strands in NGS is fragmented, and the bases in each fragment are identified by emitted signals when the fragments are ligated against a template strand. In sangers sequencing, different steps are required for sequencing, separation, and detection, which made it difficult to automate the sample preparation and it limited the throughput, resolution, and scalability. While the NGS method uses the array-based sequencing which combines the techniques developed in sanger sequencing to process millions of reactions parallelly, resulting in high throughput at lower cost.
